                                                                                               ABSTRACT

Formulation of wound-healing chitosan biomembrane has been performed. The aim of the research was study the influence ofchitosan solution on the elongation and tensile strength characteristic of the biomembrane. The degree of deacetylation and the molecular weight were determined using IR spectroscopy and viscometry methode. Chitosan 4% solution within lactic acid 2% was formulated as biomembrane with honey, PVA, and glycerol were 10, 5, and 4% respectively. This formula was divided into 6 based on the volume ofchitosan 4% solution 20, 25, 30, 35, 40% with honey and 40% without honey. The elongation and tensile strength of biomembrane were evaluated by universal testing machine and pH of biomembrane by pH meter. Results showed that the degree of deacetylation and molecular weight ofchitosan were 72.93% and 788805 respectively. The biomembranes have good elongation and tensile strength. pH of biomembranes were 4.68±0.01-5.30±0.06. Its were stable in stored at cold temperature (5 °C) and became unstable after being stored for 3 days in ambient temperature. The concentration of chitosan solution did not significant influence the elongation and tensile strength of the biomembrane statistically (P>0.05).

ABSTRAK

Formulasi biomembran kitosan penutup luka telah dilakukan. Tujuan penelitian dilakukan, untuk melihat pengaruh jumlah larutan kitosan terhadap sifat elongasi dan kekuatan regang biomembran. Derajat deasetilasi dan berat molekul kitosan ditentukan menggunakan metode spektroskopi IR dan viskometri. Larutan kitosan 4% dalam asam laktat 2% diformulasi menjadi biomembran dengan madu, PVA dan gliserol secara berurut, 10, 5, dan 4%. Formula ini dikelompokkan menjadi 6 jenis formula dengan persentase larutan kitosan 4% sebesar 20, 25, 30, 35, 40% dengan madu dan 40% tanpa madu. Bioembran dievaluasi menggunakan Universal Testing Machine untuk menentukan elongasi dan kekuatan regang, pH meter untuk menentukan pH biomembran. Hasil penelitian menunjukkan, kitosan yang diperoleh memiliki derajat deasetilasi dan berat molekul 72.93% dan 788805. Biomembran yang terbentuk memiliki elongasi dan kekuatan regang yang baik. pH biomembran 4,68±0,01-5,30±0,06. Stabil disimpan pada pada suhu dingin (5 °C) dan tidak stabil setelah hari ketiga penyimpanan pada suhu kamar. Analisa statistik menunjukkan perbedaan konsentrasi larutan kitosan tidak mempengaruhi secara nyata elongasi dan kekuatan regang biomembran (p>0.05).
